Abstract

In these studies, different serological tests were used for detection and determination of antibodies against BHV -1 in cow sera, as well as isolation of BHV -1 from vaginal discharge, nasal discharge and from aborted foeti.
These serological tests were SPA-slide agglutination test, ELISA, AGPT, FAT, VNT and SNT.
Staphylococcal protein A was prepared locally, also hyper immune serum against BHV -1 was prepared in rabbits.
Rabbits were used for studying the effect of BHV -1 rabbits at different stages of pregnancy either early (12 days) or late (25 days) non pregnancy as well as uses of different routes of infection (intranasal and intravenous) injections.
The results obtained from the present studies revealed that:
- SPA slide agglutination test revealed that 154 out of 791 serum
samples (19.5%) showed different degree of positivity
- VNT showed positivity less than that obtained by SPA agglutination test, the results were 138 (17.4%) positive VNT out of 791 serum samples.
- Indirect ELISA ”species specific” ant bovine conjugated with peroxidise revealed that 210 out of 791 (=’6.5%) showed positivity more than that obtained by VNT, while non species specific ”SPA¬ELISA” showed positive results in 187 (23.6%) less than that observed in species specific ELISA. - AGPT showed different line of precipitation in different tested serum samples as 124 samples were positive in (15,60/0), while when tested by IF AT the results of positivity were raised to reach 131 samples in 16,50/0 more than that obtained by AGPT.
The preparation of hyper immune serum against SHY -1 in rabbits showed highly positivity titters at the beginning of the 5th week post inoculation, the frequency of antibody titters were different from rabbit to another mainly at the 1 sty three weeks post injections,
- Experimentally infected rabbits by BHV -1 with intranasal instillation (UN) showed respiratory symptoms varied from mild to severe with nasal discharges, but without vaginal discharges as well as there no cases of abortions or deaths.
- The symptoms observed in rabbits infected by intravenous injection with BHV -1 were highly differentiated than that observed in cases of UN instillation that it is accompanied by abortions and sudden deaths, that 5 out of 6 pregnant rabbits were aborted between 24 hours and 5 days, while 2 out of 3 non pregnant rabbits showed respiratory and genital manifestations with deaths in one rabbit .within 24 hours post inoculation, This is means that intravenous injections of rabbits with BHV -1 is highly vigorous than intranasal instillation.
- The isolation of BHV -Ion MDBK tissue culture were obtained from 54 nasal swabs, 19 vaginal discharge and 15 aborted cow foeti, as well as resolution of virus from samples obtained from experimentally infected rabbits and their aborted foeti.
- On studying the growth curve kinetics of BHV -1, the obtained results revealed that BHV -1 shows clearly rapid growth in MDBK cells. The
