Abstract

The present investigation was an attempt to study the effect of active immunization of inunature male rabbits against inhibin on body weights and on some hormonal and biochemical parameters and on testicular biometery. Twenty immature male New Zealand White rabbits were allocated into two equal groups ten rabbits each. One group was kept as a control while the other was the treated group. Rabbits in the treated group were immunized against inhibin hormone (inhibin was partially purified from bovine follicular fluid then 0.5 mg was dissolved in 0.5 ml saline and emulsified in 0.5 ml Freund’s complete adjuvant subcutaneously injected on days 1, 14, 28 and 42. Rabbits in the control group were injected with 0.5 ml saline emulsified in 0.5 ml Freund’s complete adjuvant subcutaneously. At the 56th day, treated rabbits were injected I.V. with a booster dose of inhibin (0.5 ml saline + 0.5mg inhibin), while the controls were injected IV. with 0.5 ml saline. One week after the booster dose, sera were separated and kept frozen at - 20 °C until being used. Testis, epididymis and epididymal fat were dissected free, weighed to the nearest mg. One testis was kept frozen under -20 °C until used for some enzymes assays and zinc estimation, while the other was kept in formalin for histological examination. Serum was assayed for aspartate aminotransferase, alanine aminotransferase, blood urea nitrogen,. creatinine, total protein and albumin. Also, serum globulin and albumin- globulin ratio were estimated. Serum androstenedione and testosterone were assayed by means of radioiinmunoassay. The results revealed the following: 1. Testicular, epididymal and epididymal fat weights were significantly decreased in treated group than those of control one. 2. Testicular diameter was significantly decreased in treated group as compared to control values.3. Weekly body weights were significantly decreased in treated rabbits as compared with that of control, but there was insignificant difference in the weekly body weight gain between treated rabbits and control from the 4th to the 7th week of the experiment. <4. Active immunization against inhibin caused a highly significant decrease in AP and LDH activities and a significant decrease in zinc content of the testicular tissue in treated than that of control rabbits. 5. Active immunization caused an impairment in testicular function which appeared histologically in the form of small underdeveloped hypoplastic, distorted, closed and inactive immature seminiferous tubules which appeared with multiple spermatogonial layers. Also, the diameter of seminiferous tubules was significantly decreased compared to those of control. 6. Active inununization of rabbits against inhibin caused a significant decrease in serum testosterone and androstenedione levels as compared with those of the controls.
