Abstract WSSV is a viral infection of penaeid shrimp. The disease is highly lethal and contagious, killing shrimps quickly. Outbreaks of this disease have wiped out within a few days the entire populations shrimp farms. The presence or absence of the virus in Egyptian coast was the aim of this study. To achieve these goals, different shrimp samples from symptomatic diseased shrimp were collected from different localities at Egyptian seas and tested for detection of the virus. For isolation of the virus the collected samples of penaeid shrimp were inoculated in Vero, MDBK and MDBK supplemented with TN cell lines as mammalian cells and passaged for three blind passages. The results showed that, the percentage of the positive results after 3rd passage, of360 examined samples collected from symptomatic diseased Penaeid 16, 18, 22 samples showed clear CPE with percent of 4.44%, 5%, 6.11 % respectively. The MDBK was more sensitive than Vero cells in the isolation of the virus that was due to the first one was more acceptant for the prepared sample buffer. That . IS the cause of using MDBK with the supplementation of TN buffer. The prepared harvest solution from the different inoculated cell lines was used as templates in the amplification for PCR detection method. The obtained esults showed that, percentage of the positive PCR results from Vero cell line. |
