Abstract
Three hundred and twenty naturally infected potato plants were collected from , eight locations in Ismailia governorate , Faid [Srapuom , Abu –Sultan ] , Abu -Swear [Saba- Abar , El-Mahsama , Abu –Swear station ] , El-Kantara garb [ Riad , El- Blah ] and El –Tall [El-Tall –El-Caber ]. The original PLRV isolate was obtained from potato plants [Collected from El- Tall El-Caber location] which showing the identical PLRV virus symptoms. Samples were inspected for PLRV symptoms and indirect ELISA was used to confirm the presence of PLRV infections. All tested plants collected from El- Tall El-Caber farms were gave positive reaction with specific PLRV antibodies by indirect ELISA.
This isolate was used for virus characterization and its effect of different cultivars of potato plants. According to host range, differential hosts, symptomatology, virus stability, insect, tuber and grafting transmission, inclusion bodies, electron microscopy and serological reaction. Identification was performed as the following:-
Fifty – seven plant species and cultivars belong to sex different families were inoculated with PLRV using infective green peach aphids Myzus persicae Sulze , Symptoms appeared on tested plants were recorded daily nearly for one month after inoculation . These results demonstrated that:-
(I) Susceptible hosts:
Solanum tuberosum CVs. Spunta , Nicola , Diamont, Alpha , Draga and Kara Datura stramonium L. , Datura tatula L., Physalis floridana Rydb., Physalis peruviana L. and Lycopersicum esculentum CVS. Brigade , Super sierra and Kastslia .
(II) The carrier hosts:
Nicotiana glutinosa , Nicotiana tabacum , Nicotiana clevelandii , Datura metal and Capsicum annum CVs . Lamuyo, Sonar, Pical and Bruyo .Plants by means of aphids showed that these hosts carried the PLRV, which gave +ve by indirect ELISA.
(III) The non-susceptible hosts:
Chenopodium amaranticolor ,Ch. quinoa ,Ch.album , Vicia faba L. ,V. narbonensis , V. cerenia , Phaselous vulgaris L. , Glycine max L. , Vigna sinensis L. , Pisum sativum L. , Arachis hypoyaea L. ,Cucurpito pepo , Cucumis sativus , Seramum indicum ,Brassica oleracea CVs. Botrytis and B. Oleraceae CVs. Capitata and B. rapa . , which gave –ve by indirect ELISA .
