Abstract One hundred and sixty albino rats were divided into eight groups as follow: Group A: This group contains 20 rats. The rats were injected with diclofenac sodium daily with a dose of 1.35 mg/rat/day. Samples of blood and organs were collected from 5 rats every week for four weeks. Group C-A: This group contains 20 rats. The rats were injected with saline daily as a control group. Samples of blood and organs were collected from 5 rats every week for four weeks. Group B: This group contains 20 rats. The rats were locally applied with diclofenac diethylammonium daily. Samples of blood and organs were collected from 5 rats every week for four weeks. Group C-B: This group contains 20 rats. The rats were locally applied with emulgel daily as a control group. Samples of blood and organs were collected from 5 rats every week for four weeks. Group C: This group contains 20 rats. The rats were injected with diclofenac sodium twice weekly with a dose of 1.35 mg/rat/dose. Samples of blood and organs were collected from 5 rats every two weeks for eight weeks. Group C-C: This group contains 20 rats. The rats were injected with saline twice weekly as a control group. Samples of blood and organs were collected from 5 rats every two weeks for eight weeks. Group D: This group contains 20 rats. The rats were locally applied with diclofenac diethylammonium twice weekly. Samples of blood and organs were collected from 5 rats every two weeks for eight weeks. Group C-D: This group contains 20 rats. The rats were locally applied with emulgel twice weekly as a control group. Samples of blood and organs were collected from 5 rats every two weeks for eight weeks. Blood samples were collected from the retro-orbital venous plexus of rats for haemogram and serum biochemical analysis. The haematological findings showed that there were decrease in RBCs count, Hb concentration and PCV. The blood indices show normocytic normochromic red blood corpuscles that indicate aplastic anaemia due to the effect of the drugs on the bone marrow and it is supported by the presence of megakaryocytes in the spleen as a mark for extramedullary haematopieosis. There were decrease in number ofWBCs and also there were increase in the number of lymphocytes and monocytes that mean agranulocytosis. The clinicopathological results showed hepalocellular damage manifested by increase in AST, ALT , total, direct and indirect bilirubin ” . |
