Abstract

In the present study, we determined the in vitro cytochrome P450-
mediated biotransformation activities for five pharmacologically
important substrates in male leghorn chickens and beagle dogs to evaluate
the liver catalytic activities in normal chicken and compare it with those
of dogs. According to our results, the chicken total cytochrome P450
content (0.296±0.019 nmol/mg protein) was almost similar to the values
reported in humans, cats, pigs, trout fish, snakes, and frogs. On the other
hand, chicken total cytochrome P450 content was lower than levels
reported for most of the other species including the beagle dog
(l.114±0.099 nmol/mg protein). We also investigated the effects of
lipopolysaccharide on the cytochrome P450-mediated activities and
antipyrine pharmacokinetic to evaluate the drug catalytic activity in
diseased chicken. After injection of lipopolysaccharide (50 mg/kg b. w.)
to chicken, total hepatic microsomal cytochrome P450 content was
decreased by about 20%, antipyrine elimination rat constant was also
decreased by about 44%, while antipyrine plasma half-life was
significantly increased by about 77%.
