Abstract

In this study, three types of vaccines were prepared. K45/20A Brucella oil adjuvenated
(Montanide ISA25) vaccine, BEF-BEI inactivated and adjuvenated with (Montanide ISA25)
and the third one of vaccine was a combined one which contained both types of organisms
(Brucella & BEFV). Each type of vaccine was used in vaccination of a group of calves (four
calves in each group with two unvaccinated controls). The humoral immune response against
Brucella abortus vaccines was determined by three techniques, rose bengal test (RBT), tube
agglutination test (TAT) and complement fixation test (eFT) and by using delayed
hypersensitivity test for cellular immunity. On the other hand, the humoral immune response
against BEF vaccine was evaluated by serum neutralization test (SNT) and enzyme linked
immunosorbent assay (ELISA) and by using lymphocyte transformation or blastogenesis test
for cellular immunity. The results showed that using of Brucella K45/20A-BEF combined
vaccine increase the immunity generated when BEF inactivated vaccine is used singly, while
the picture was not changed too much when K45/20A vaccine used alone or combined with
BEF. Montanide ISA25 is a mineral oil, used as an adjuvant for the first time in the
preparation of Br K45/20A vaccine, BEF inactivated vaccine and the combined vaccine.
So, it is recommended to vaccinate the Egyptian cattle with the combined vaccine which
increase the BEF vaccine immunity with a conserved Brucella immunity, saving effort, time
and money.
