Abstract

Abstract Cases of cattle infected with bovine ephemeral fever virus were suspected in 7 governerorates (Damietta, Al sharquia, Al fayoum, Elmenoufia Alexandria, Suez and Behira ) the suspected samples were collected in summer season in 2006 and 2009 the total number of samples were 115 serum samples for antibody detection, 95 blood with anticoagulant and 206 nasal swabs for virus isolation. All samples were collected from small stalk held having small number of animals and from dairy farm in damietta . The virus was isolated in VERO cell line the results were0% from nasal swabs and 17.8 % from Buffy coat while the isolation in 2 days old baby mice I/C inoculation were 0% from nasal swabs and 23.1 from Buffy coat. Also identication of the viral isolates were conducted by indirect immunoflourcent technique showed positive result 36.6 % from buffy coat while the confirmatory results of positive by reverse transcriptase polymerases chain reaction for identification of the virus showed a clear single band on agarose gel stained with ethidium bromide The amplified RNA fragment corresponds to 500 base pair (bp) in ratio 45.4% from buffy coat. Serum neutralization test was used for screening of antibody to the virus and the positive result, were 36.5 % and the titre ranged between 4 to 64
