Abstract This study was conducted from ٢٠٠٢ through ٢٠٠٨ using both semi dry date palm cultivars i.e., Siwy, Oshkingbeel and dry date palm cultivars i.e., Fraihy and Ke’aby. The study comprised two parts: Part I: In vitro propagation of the four cultivars via somatic embryogenesis and study the genetic stability between the mother plant and the produced plants from micropropagation using RAPD analysis; -Both shoot tip and leaf primordia explants of date palm Siwy, Oshkingbeel, Fraihy and Ke’aby cvs. were successfully disinfected using different disinfection methods. The highest survival percentages for both explant type were achieved through multi- step treatment using ٢,٦ % NaOCl for ٣٠ min followed by ٢,١ % NaOCl for ١٥ min followed by ٠,١ % HgCl٢ for ٧ min. shoot tip gave the highest significant survival percentage.- November was the most suitable time for explant preparation, which gave the lowest browning percentage, -MS culture medium supplemented with ١٠٠ mg/l ٢،٤-D+٣ mg/l ٢iP was effective on swelling percentage.-Shoot tip explants of Siwy, Oshkingbeel, Fraihy and Ke’aby cultivars proved to be the most suitable explant for starting and initiation of callus cultures, followed by leaf primordial explants. |
