Abstract

The characterization and Phylogenetic analysis of RVF-ZHsOl strain used in vaccine production were not studied yet. So, this study aimed to identify the vaccinal strain. Firstly, the RVF virus was purified using plaque purification technique, the major equally-sized plaques (large plaques) were picked, then RNA of Gn gene of M segment was extracted and ampilified by RT-PCR. Electrophoresis ofRT-PCR product was done. The DNA band was sliced from the gel and purified followed by sequencing. The resultant sequence was aligned with those of previously characterized RVFVs by using Bioedit program which revealed five nucleotide substitution and lead to three mutations in translating amino acid. So, this study put an end to the accusation of RVF vaccinal strain m any commg outbreak as well as revealed the identification of this vaccinal strain.
