Abstract Infertility in buffaloes is considered as one of the most important disorders which affects on the productivity and reproductivity. The disease under concern in this study is Brucellosis in Buffaloes. 102 blood samples and 17 tissue samples were obtained from Buffalo cows from Governmental farm located at Ismaillia govemorate. 15 tissue samples were obtained from obligatory slaughtered cases from Ossim slaughtered house in Giza governorate. All samples were examined for brucellosis by using different diagnostic methods including serological tests, isolation of Brucella organism, gross and histopathological examination in the reproductive and lymph reticular system, detection of Brucella antigen in tissue by immunohistochemistry and finally detection of Brucella DNA in tissue by Pf’R. The obtained results of serological tests were 16.6 0/0, 1l. 7 6 %, 12.74 %, 1l.76 % and 13.72 % with RBPT, TAT, MET, CFT and ELISA respectively. Brucella was isolated from three out of thirty two examined cases (9.37%). The most pronounced histopathological changes were granulomatous, ulcerative and hemorrhagic endometritis as well as granulomatous mastitis. Moderate to severe immuno-reactive staining of brucella antigen were detected in supramanunary lymph node, spleen and uterus. PCR assay of uterus and supramammary lymph node gave positive PCR product (223bp) and (731 bp) using of Brucella species primers and Brucella melitensis primers. |
