Abstract

Abstract In the present study, bacteriological examination of 100 semen samples(cattle, buffalo and human) was carried out. Only 22 samples were positive for E. coli, while all the straw samples came negative for E. coli isolation. Serotyping revealed E. coli 06. The hemolytic activity of the isolates showed a type of hemolysis on ’sheep, cow, and human blood agar. Hemagglutination pattern and their relation to suspected CFA of Isolated strains was determined. By incubation of E. coli 06 with separated sperm, sperms were highly agglutinated with low viability. The sperm- E. coli 06 agglutination was blocked using different treatments (0- mannose, different antibiotics and specific E. coli hyperimmune serum). ’ Results confirmed by IVF experiments in ’buffalo. When, TCM-199 medium was used. And separated motile sperm or addition of specillc E. coli 06 hyperimmune serum to buffalo sperm when, compared with addition of E. coli 06 or specific E. coli 06 hyperimmune serum + E. coli 06.: Results included: increase of sperm agglutination with decrease of sperm viability due to E. coli 06 with reduced IVF and embryo production results. Whe buffalo spermatozoa incubated with speCific E. colf hyperimmune and E. coli 06 before fertilization. The improvement effect of specific E. coli hyperimmune setum on the fertilization, cleavage and transferable embryo rates was clear in comparison to the effect of E. coli 06 and control experiments.
