Abstract A total of 360 serum samples, 20 Buffy coat, 20 saliva and 20 epithelial tissues were collected from cattle from different localities of Assiut Governorate (Arab El- Awamer, Sahel seleem, Dairoot, Veterinary hospital, Aboteeg, Garb elbalad and Assiut) were used in this study and subjected to different epidemiological, serological and virological studies as summarized below: 1. The seroprevalence rate of FMDV NSP (non- structural protein) antibodies in examined cattle at Assiut Governorate revealed that the infection rate was 42.2% (Arab El- Awamer 47.4%, Sahel seleem 33.3%, Dairoot 18.2%, Veterinary hospital 25%, Aboteeg 20%, Garb elbalad 27.8% and Assiut 26.3%). 2. Clinical examination of infected animals infected with FMDV revealed fever, excessive salivation, stomatitis, and lesions in feet and in dairy cattle, vesicles detected on the udders. 3. Female cattle were more susceptible than males to infection with FMD (female 65.8% (100/ 308) - males 34.2% (52/ 52)). 4. The infection rate was 88.2% in Old cattle (Above 4 years), 9.9% in Adult (Between 2 and 4 years) and 2% in Young (< 2 years). 5. Higher rate of infection was in non- hot months 47.03% than hot months 33.06%. 6. Serum neutralization test was used for detection of FMDV serotypes O and A antibodies in cattle sera, it was found that the total positive sera for FMDV serotype O were 236 (65.6%) out of 360 examined sera. While it was found that the total positive sera for FMDV serotype A were 214 (59.4%) out of 360 examined cattle sera. 7. Liquid phase blocking ELISA was used for detection of FMDV serotypes O and A antibodies in cattle sera, it was found that the total positive sera for FMDV serotype O were 270 ( 75%) out of 360 examined sera. While It was found that the total positive sera for FMDV serotype A were 253 (70.3%) out of 360 examined sera. 8. PrioCHEKIT FMD-ELISA was used for detection of nonstructural protein antibodies against FMDV. It was found that out of 360 examined sera 152 (42.2%) were positive for the presence of nonstructural protein antibodies against FMDV detected in cattle. 9. Trials for Isolation of suspected FMDV from Buffy coat, epithelial tissue, and saliva collected from infected cattle by three blind serial passages on BHK21 cell culture. It was found that the positive samples induced cytopathic effect after 24-48 hours 10. Indirect sandwich Enzyme Linked Immunosorbent Assay (ELISA) was used for Identification and serotyping of field isolates of suspected FMDV showed that type O and A were detected in the examined samples by percentage of 27(67.5%) out of 40 and 23 (57.5%) out of 40 respectively. 11.
