Abstract The aim of the present work is to investigate the effects of terfenadine and astemizole on some cardiovascular parameters; on the inotropic and chronotropic activities and coronary outflow of isolated rappit hearts (in vitro study), on and also, on the mean arterial blood pressure, heart, PR interval and rate-corrected QT (QTc) interval in the anaesthetised rats (in vivo study). In addition, this study was done to evaluate the effects of terfenadine or astemizole, on these cardiovascular parameters, in the presence of amiodarone or hydrocortisone in the anaesthetised rats. Amiodarone was selected because it is widely available and Frequently used as a class III antiarrhythmic agent, with known potential cardiac arrhythmic toxicity (e.g., prolonged QT interval and rare cases of torsades de pointes). while, hydrocortisone was selected because it is widely available and frequently used for similar indications of terfenadine and astemizole, and it could be used in combination with either of the two non-sedating antihistamines. |
