Abstract

Four isolates of spodopetra littoralis suspected to be Infected by nuclear polyhedrosis (SpLiMNPN) virus had been collected from different governance (Cairo, Fayoom, Qalubiya and Monofeya) and tested for infection using polymerase chain reaction by designed primer based on published sequencesfor polyhedrin gene of SpLiMNPV.
Infected larvae used for viral purification and multiplication in the viral host spodopetra littoralis. potency of infection tested and the result indicate that the Cairo isolate is highest mortality number at th 3rd and 5th instar of larvae.
Nine isolte of serratia marssence tested for Chitinase activity using the clearing zone test on chitin plate and the highest chitinase activity strain was S1, S3, S2 and S12 respectively. PCR amplification of ChitinaseB had been done for the nine isolates by designed primer based on published sequences for testing the existence of ChitinaseB gene, and the PCR product of isolate S1 used in viral transformation construct.
Viral transformation construct designed by Discovery studio gene software and constructed by tow steps. Firstly polyhedrin gene cloned to pBK-CMV vector and selected by white and blue colony selection, secondly the selected and tested colonies used for plasmid extraction and cloning of ChitinaseB gene and the selection at this step performed using clearing zone test.
