Abstract

The present work was carried out to evaluate the advantages of ozone therapy in hepatocellular carcinoma in rats.
The study was performed on 64 albino rats. These rats were divided into four groups:-
 Control group (C-group): included 10 rats which didn’t receive treatment or ozone doses.
 Ozonized control group (Ozonized C-group): included 10 rats. Ozone in oxygen by rectal insufflations 0.7mg/kg - five doses per week were given for three weeks one session per day.
 (DBA-group): included 22 rats. Rats were kept for 9 months fed with bread in milk and 2500 ppm of sodium nitrate and 1250 ppm dibutyl amine (DBA) dissolved in drinking water with no limit of drinking volume.
 (Ozonized DBA-group): included 22 rats. Rats were incubated for nine months fed with bread in milk and 2500 ppm of sodium nitrate and 1250 ppm dibutyl amine (DBA) dissolved in drinking water with no limit of drinking volume. Rats received ozone in oxygen by rectal insufflations 0.7 mg/kg - five doses per week for three weeks.
A piece was taken from each rat liver tissue for histopathology Many tests were included in this study , biochemical measurements includes [liver function tests (SGPT, SGOT and Alkaline Phosphatase) and oxidative stress (Catalase, Nitric Oxide and Malondialdehyde)] and physical measurements (pH value and conductivity).
All results obtained were statistically analyzed using statistical package for social sciences (SPSS) analysis. The results obtained reveled that:
Liver function tests:
 In (DBA-group) the mean value for Alkaline Phosphatase it was increased by 41 % compared to its value in the control animals (C-group), the (SGOT) mean value when compared to the (C-group) the increasing was of 34.9% ,and ( SGPT) increased by 31% than (C-group).
 In the (Ozonized DBA-group) serum samples revealed that the mean value of Alkaline Phosphatase level was reduced by 41% compared to its mean value in the (DBA-group) and SGPT levels mean value in the (Ozonized DBA-group) was slightly decreased by 11.29% than its mean value in (DBA-group). There was a significant decreasing in SGOT mean value in (Ozonized DBA-group) comparing to its value in (DBA-group) by 16.4%.
 In the (Ozonized C-group), by studying the effect of ozone doses on the (C-group) readings it was recorded both of SGOT and SGPT mean value decreased in a reduction ratio of 39.47% and 68.1% respectively. Alkaline phosphatase mean value reduced by 21.7% from in the (Ozonized C-group) but in non significant change.
Oxidative stress tests:
 The (MDA) mean value in the (C-group) was raised in (DBA-group) by increasing ratio 16.58%. The (NO) mean value in (C-group) was raised by 10.59% in (DBA-group). The antioxidant enzyme Catalase, its mean value was increased from in the (DBA-group) by 20.46% than its value in (C-group).
 The (MDA), (NO) and Catalase mean value in the (Ozonized DBA-group) was raised by 30.4% , 49.7% and 20.3% respectively than in the (DBA-group).
 The change in the MDA level mean value increased by 86.1% in the (Ozonized C-group) than in the (C-group), the mean values of NO and Catalase increased by 10.59% and 19.69% in the (Ozonized C-group) than in the(C-group).
Biophysical tests:
 In biophysical tests There was a significant deceased in the pH value and conductivity of rat’s blood noticed by the biophysical data for (DBA-group) in comparison to (C-group). The mean value of pH in (DBA-group) was reduced by 7.24% than that in the (C-group) and the conductivity mean value was reduced in (DBA-group) by 13.1% than its value in (C-group) .
 In the (Ozonized DBA-group) the mean value of pH in was the same of that in the (DBA-group) and the mean value of conductivity has a slight reducing than the (DBA-group) in a non significant results.
 The resulted change in the mean values for pH and conductivity by ozone doses in (Ozonized C-group) were highly significant where the pH mean value reduced by 11.59% from the (C-group) to the (Ozonized C-group), and the mean value for conductivity was decreased by 14.75% from the (C-group) to the (Ozonized C-group).
 There were a highly significant reduction in the pH mean value by 7.2% from the (C-group) to the (Ozonized DBA-group). The Conductivity mean value was reduced in the in the (Ozonized DBA-group) by 13.4% than in the(C-group).
Histopathological results:
 In the (DBA-group), the histopathological examination showed hepatocytes with foamy cytoplasm and vesicular nuclei, congested central vein surrounded by microvesicular and macrovesicular lipidosis and, slight hemorrhage in the blood sinusoid; peripheral lobular necrosis and hemorrhagic areas; lymphocytic cell infiltration; congested blood vessels through extended necrosis and pronounced degenerative areas ; hyperplasia of epithelial lining bile duct and sever hemorrhage of blood capillary ; vacuolar degeneration of hepatocytes ; foci of necrotic hepatocytes as well as portal infiltration with lymphocytes and lymphocytic cells infiltration in the portal triad associated with ballooning of the hepatocytes.
 In the (Ozonized DBA-group), Ozone treatment improved towards normal the liver structure. There was only few hemorrhagic areas and slight foamy appearance of hepatocytes and slight hyDROPic degeneration of hepatocytes occurred . In addition, there was mild kupffer cells activation with only few hemorrhagic areas and clearly appeared normal central vein. However there was wonderful healing effect showing apparent normal hepatocytes strands.
