Abstract

This thesis is devoted to the analysis of some second generation H1 receptor antagonist drugs; namely fexofenadine hydrochloride, ebastine, and cetirizine dihydrochloride. A simple and sensitive first and second-derivative spectrophotometric and highly sensitive first and second-derivative synchronous spectrofluorimetric method for the simultaneous analysis of fexofenadine hydrochloride in presence of its different degradation products was developed. The proposed method was also subjected to validation and to detailed kinetic study. A reversed phase liquid chromatographic method for the determination of the same drug in the presence of its forced degradation products is presented; the method was utilized to investigate the kinetics of the different degradation products of fexofenadine, separate them and conclude different degradation pathways of the studied drug. A sensitive spectrofluorimetric method for the determination of ebastine in pharmaceuticals depending on condensation with mixed anhydrides producing a product with intense fluorescence was developed. The method was utilized to investigate the kinetics of the degradation of the drug. The thesis describes a sensitive spectrofluorimetric method for the determination of ebastine in pharmaceuticals depending on quantitative fluorescence quenching of eosin which was extended to apply first and second derivative synchronous spectrofluorimetric method for the simultaneous analysis of the drug in presence of its degradation products. Another part is concerned with the development of an accurate, simple, sensitive and selective reversed phase liquid chromatographic method for the determination of EBS in its pharmaceuticals after pre column derivatization with Zn2+ ions. The composition of the complex as well as its stability constant was also investigated. Moreover, the proposed method was found to be a stability indicating one. In another part, an accurate, simple, sensitive and selective micellar liquid chromatographic method is introduced for the simultaneous determination of cetirizine dihydrochloride in two of its combined pharmaceutical preparations with psudoephedrine hydrochloride and /or paracetamol. The chemometrics approach was applied for the optimization of separation of the studied drugs by means of multivariate analysis. The method was applied for the determination of the drugs in spiked human plasma, and used to reveal their pharmacokinetic characters. Another part presents a validated simple, economic, selective, and stability indicating spectrofluorimetric method for the determination of ebastine, cetirizine dihydrochloride, and fexofenadine hydrochloride based on their reaction with 2-cyanoacetamide in alkaline medium to give highly fluorescent derivatives. The method was applied for the determination of the drugs in spiked human plasma, and used to reveal their pharmacokinetic characters. The method was also utilized to investigate the kinetics of different degradation products of the drugs. The last part aims to develop a validated simple, rapid, and selective spectrofluorimetric method for the determination of FXD, EBS, and CTZ based on the reaction of the cited drugs with some Π acceptors namely p-chloranilic acid, tetracyanoethylene, and 2,3-dichloro-5,6-dicyano-p-benzoquinone to give highly fluorescent derivatives. Reactions Stoichiometries of the complexes were also investigated.
