Abstract In the present study, comparison of some virulence factors and resistance between staphylococci isolated from diabetic and non-diabetic patients was done. Eighty six microbial isolates were separated and identified as S. aureus (58 isolates) and CoNS (28 isolates). The resistance patterns of all staphylococci isolates were determined against 8 antimicrobial agents. Detection of some virulence factors among tested staphylococcal isolates was performed using phenotypic methods. These virulence factors include hemolysins, lipase and lecithinase enzymes, haemagglutination, biofilm formation and carotenoid pigment production. The virulence genes hla, hlb, icaA, icaD, geh and methicillin resistance gene mecA were detected by PCR on total DNA of S.aureus isolates. Nucleotide sequencing of hla, icaA and mecA genes was carried out for 10 isolates of S.aureus using modified sanger sequencing technique. Also, this study tested the effect of sub-inhibitory concentrations of clindamycin, amikacin and vancomycin on α-hemolysin production and spa gene expression in PC203 reporter strain. |
