Abstract

Results of studying production of bioactive compounds from in vitro grown seedlings showed that in vitro grown seedlings at different growth stages gave amounts of total soluble proteins, total carbohydrates & total cardiac glycosides mg/g DW higher than the wild plant. Total cardiac glycosides & total soluble protein from dried latex of wild plants was higher than from in vitro grown seedlings.
Testing the effect of temperature (a physical factor) and the effect of thiamine & pyridoxine (a chemical factor) on production of both total proteins and total carbohydrates of suspension cultures results showed that total carbohydrates and total proteins concentration from suspension cultures for both tested factors was higher from hypocotyls than from cotyledonary leaves suspension cultures.
Cell suspension cultures of both hypocotyls and cotyledonary leaves of medicinal plant Calotropis procera were used for studying a number of factors affecting the production of cardiac glycosides & the growth of cells. The study resulted in identifying significant differences for all the studied factors on the production of cardiac glycosides in cell suspension cultures.
Using different inoculum densities showed that a 0.5 g inoculum density gave highest concentration of cardiac glycosides of 3.38 and 3.37 mg/g DW for hypocotyls and cotyledonary leaves respectively.
Studying different concentrations of sucrose revealed that 7.5% sucrose gave highest mean cardiac glycosides for both ex plants of 9.06 mg/g & 2.38 mg/g for hypocotyls & cotyledonary leaves respectively.
The study result in identifying the best pH value, media strength, temperature, the type of sugar, concentration of thiamine & concentration of pyridoxine for growth of cells (fresh & dry weight) & cardiac glycosides concentration.
