Abstract irology.This work was carried out for molecular comparison between FMDV field strains causing recent outbreaks in Egypt and the used vaccine strains. 252 epithelial samples and 13 8 oesophageal pharyngeal fluids were collected from 9 different Egyptian governorates during the period of2009 to 2012. The result obtained from this study can be summarized as followings: . 1. Isolation and identification of the FMDV current field strains by tissue culture inoculation, baby mice infection and by R T- PCR using universal primer. The collected samples were isolated in BHK cell line for three successive passages and also by baby mice inoculation observed for seven days, all the suspected samples are positive. The positive samples then retested by RT-PCR for general detection of FMDV using universal primer, all samples are positive. Typing of the isolated FMDV strains by CFT using specific guinea pig antisera to the 146S antigen of each of the seven serotypes of FMDV, ELISA using specific rabbit antiserum as a trapping antibody, and specific guinea pig antisera as the detecting antibody, and RT-PCR using RNA extraction by GeneJET™ RNA Purification Kit (Fermentas, EU), RT-PCR by Maxime” RT-PCR PreMix Kit (iNtRON, Koria) and five |
