Abstract

Cadmium and lead are highly toxic metals. People are exposed to them primarily through food and water. Pomegranate seeds and peel are considered good source of flavonoids, polyphenols, pectin and carotenoids which have antioxidant potential. Therefore, the aim of the research was to study the using of pomegranate against toxicity induced by contaminated diet with lead and cadmium toxicity in rats. Pomegranate seeds and peel were dehydrated and evaluated chemically by compound (as polyphenols, carotenoids, total flavonoids and anthocyanine), some polyphenols fractions, and pectin. Forty five albino male rats (Sprague Dowely strain) of an average weight 130 ± 10 g were divided into 9 groups each group contains 5 rats. G1- negative control rats fed on basal diet, G2- positive control rats received (contaminated diet with lead),G3- positive control rats received (contaminated diet with cadmium),G4- positive control rats received (contaminated diet with lead and cadmium) G5 - rats received (contaminated diet with lead and 5% dried pomegranate seeds) G6- rats received (contaminated diet with lead and 5% dried pomegranate peel)G7- rats received (contaminated diet with cadmium and 5% dried pomegranate seeds),G8- rats received (contaminated diet with cadmium and 5% dried pomegranate peel) G9- rats received (contaminated diet with lead and cadmium + 2.5% dried pomegranate seeds+2.5% dried pomegranate peel) for 5 weeks. The results revealed that positive control groups gave a very significant increased in liver functions ”alanine aminotransferase (ALT), aspartate aminotransferase (AST), Alkaline Phosphatase (ALP) activities”, kidney functions (creatinine and urea); and total cholesterol (TC), triglyceride VLDL-c, LDL-c. Significantly decreased in glutathione peroxidase (GPX), blood hemoglobin, HDL-c values, body weight and feed efficiency ratio.These results indicated the toxicity of lead and cadmium. However, the groups received (contaminated diet with lead and 5% dried pomegranate seeds), (contaminated diet with cadmium and 5% dried pomegranate seeds), (contaminated diet with lead and cadmium+ 2.5%dried pomegranate seeds+2.5% dried pomegranate peel) significantly improved in glutathione peroxydase, blood hemoglobin, body weight and feed efficiency ratio, lipid profile (TC, TG, HDL-c, VLDL-c, LDL-c) and ALT, AST, ALP, creatinine and urea. The results also showed that the groups received (contaminated diet with lead and 5% dried pomegranate peel), (contaminated diet with cadmium and 5% dried pomegranate peel) showed a highly significant improved, as compared to the control negative (G1) and positive control (G2, G3, G4) and around negative control (basal diet).It was noticed that the group received pomegranate peel showed the better result than pomegranate seeds. These results indicated the potential protective action of pomegranate against lead and cadmium toxicity. Also histopathological examination revealed an improvement in organs tissues. These results indicated the protective effect of pomegranate (seeds-peel) against lead and cadmium toxicity. This may be due to multiple line defenses of powerful antioxidants (polyphenols, total phenols, total carotenoids, total flavonoids, anthocyanine and phenolic compounds) and fibers which present in high levels in pomegranate.
