Abstract

This work was conducted to study some biological effects of Ezalo(R) mat aerosols which contains 40mg bio-allethrin, 20mg pyrethrum extract 25% 30mg piperonyl butoxide and 20mg butylated hydroxy toluene through haematological, biochemical and histopathological studies. Experiment was carried out on six groups, each of 13 albino rats of both sex. first, second, third and fourth (control) group were given intraperitoneal injections twice a week of 34mg/kg.b.w. bio-allethrin, 34mg/kg.b.w. bio-allethrin with 17mg/kg.b.w. piperonyl butoxide, 150mg/kg.b.w. butylated hydroxy toluene and an equivalent volume of corn oil for up to 16 successive weeks. Fifth group inhaled aerosols of one tablet of Ezalo(R) mat for 10hr/day for 16 successive weeks, while sixth group was kept as a control for fifth group. from the obtained results bio-allethrin caused non-significant difference in body weight at 2,4,6,8,10,12,14 and 16. weeks of injection, while synergized bio-allethrin caused decrease in body weight at 4,12,14 and 16 weeks. Butylated hydroxy toluene induced decrease in body weight at 6,8,10,12,14 and 16 weeks of injection, while the fifth group that inhaled Ezalo(R) aerosols showed a decrease in body weight at 6,8,14 and 16 weeks of inhalation. Bio-allethrin or synergized bio-allethrin resulted in non-significant difference in R.B.Cs count, haemoglobin concentration and haematocrit values, while butylated hydroxy toluene induced decrease in R.B.Cs count at 8,10,12 and 16 week, Hb concentration at 4,8,10 and 12 weeks, and haematocrit value at 8,10 and 12 weeks of injection. Inhalation of Ezalo(R) aerosols produced decrease in R.B.Cs count at 4,8,10 and 16 weeks, haemoglobin concentration at 4,6,8,14 and 16 weeks and haematocrit at 8,10 and 12 weeks of
inhalation. Intraperitoneal injection of bio-allethrin or butylated hydroxy toluene induced non-significant effect on white blood cell count, while intraperitoneal injection of synergized bio-allethrin caused increase in the leukocytic count at 8,10,12,14 and 16 weeks of injection. Inhalation of Ezalo(R) aerosols produced an increase in leukocytic count at 10,12,14 and 16 weeks of exposure. Bio-allethrin induced increase in lymphocytes at 4,8 and 10 weeks with decrease in neutrophils at 4,14 and 16 weeks, while synergized bio-allethrin caused increase in lymphocytes at 4,8,10,12 and 14 weeks with decrease in neutrophils at 4,12,14 and 16 weeks of injection. An increase in lymphocyte counts were observed in group injected by butylated hydroxy toluene at 8,10 and 12 weeks, while neutrophil was decreased only at 10 week of injection. Inhalation of Ezalo(R) aerosols resulted in increase in lymphocytes at 10,12,14 and 16 weeks of inhalation.
Intraperitoneal injection of bio-allethrin, synergized bio-allethrin and inhalation of Ezalo(R) aerosols induced hyperglycemia, while butylated hydroxy toluene caused hypoglycemia. Biochemical data indicated that bio-allethrin caused non-significant effect in SGPT, SGOT, alkaline phosphatase, urea, uric acid, creatinine, total protein, albumin, and potassium, while sodium concentration showed significant increase. Intraperitoneal injection of synergized bio-allethrin induced significant increase in SGOT, alkaline phosphatase, serum urea and serum potassium concentration, while high significant increase was noticed in serum Got in addition to a non-significant effect in serum uric Acid, creatinine total proteins, albumin and serum sodium
concentration. Intraperitoneal injection of butylated hydroxy toluene resulted in significant increase in serum GPT and serum sodium concentration while serum alkaline phosphatase and serum total protein showed high significant increase while serum potassium concentration showed significant decrease, in the other hand non-significant effects were noticed in the serum Got, urea, uric Acid, creatinine and albumin level. necrosis while heart revealed mononuclear cell aggregation, area of degeneration and necrosis of myocyte. Liver in all cases showed mono nuclear cell aggregation in portal area and increased percent of binucleated cells. Kidneys in many cases showed focal area of tubular basophilia and papillary epithelial cell hyperplasia. Hyperplasia of lymph follicles was evident in the spleen. Lung in many cases of the group inhaled Ezalo(R) mat showed perivascular and peribronchial cuffing of lymphocytic type, form cell aggregation, interstitial pneumonitis and edematous fluid was observed in few cases. Liver revealed degenerated hepatocyte which appear vaculated as foom, proliferation of life epithelial cells and activation of van kuffer cells. Convoluted tubule of the kidneys showed degenerated changes, hyaline cast and shrinkage glomerular tuft in scattered case. Spleen showed hypoplasia of lymph follicles.
