Abstract The present work aimed to study the β-glucosidase production and its properties to obtain maximum production by different fungi using different optimization methods under solid state fermentation on rice straw. The fungal strains were isolated from different soils, rice straw and compost samples. The most active fungi in β-glucosidase production under SSF were selected. Plackett-Burman fractional factorial design was used to screen different nutrients and physical factors and choose the most significant factors that affected the production of β-glucosidase. β-glucosidase production was optimized using response surface methodology (Box-Behnken design) in order to obtain the highest production of β-glucosidase. The enzyme was purified by gel filtration method. In addition, physiochemical and kinetic properties of this enzyme were studied. |
