Abstract

Hypercholesterolemia represents the major public health disease affecting different body organs. Both metabolic diseases affected drastically the mother during pregnancy as well as retarded the differentiation of different body organs and shared in development of different pattern of congenital abnormalities. Two hundred and fortty fertile male and virgin female rats were used for experimentation. Females were mated in a special cage (1male/3 females) during overnight and gestation was determined in the next morning by the presence of sperm in a native vaginal smear. The pregnant rats were arranged into twelve groups; control, pomegranate juice or Morus alba leaves extract, experimental hypercholesterolemic with or without either morus alba leaves extract or pomegranate juice, Lepitor-treatment with or without either morus alba leaves extract or pomegranate juice, hypercholesterolemia and lepitor with or without either morus alba leaves extract or pomegranate juice. Each of the experimental groups was sacrificed at parturition as well as at 1,7,14 and 21-d postnatal. Light and transmission electron microscopy, determination of neurotransmitters (dopamines, ?-aminoputyric acid and serotonin), biochemical markers of cell death caspase 3 & 7 , VEGF and 8-hydroxydeoxyguanosine and comet assay of DNA damage were subjected for investigation. The following parameters are investigated: 2. SDS-PAGE analysis & GC-MS mass analysis of phytochemical components of Morus alba and pomegranate. 3. Biochemical analysis of retinal and cerebellar markers. 4. Light and transmission electron microscopic investigation of retina and cerebellar cortex. 5. Assessment of retinal and cerebellum neurotransmitters. 6. Assessments of DNA fragmentation. 7. Assessments of immunochemistry of retina.
