Abstract

This study was under taken to show the effect of different doses of
gamma radiation (1, 2, 3 and 5KGy) on the metabolism of some seeds born
fungi isolated from maize and wheat seeds collected from different local
markets from El-Gharbia to control growth of more common fungi during
storage.
Nine fungi were isolated from maize and wheat seeds. The most
common species were Aspergillus terricola and Aspergillus fumigatus.
Gamma ray has no effect on maize and wheat seeds germination. Gamma
radiation leads to several mutagenic change which affected on germination,
viability and germ tube length of Aspergillus terricola and Aspergillus
fumigatus spores which decreased by increasing gamma rays. Also, gamma
irradiation affected on the dry biomass and radial growth in both fungi. The
content of total proteins, carbohydrates and lipids decreased with increasing
the irradiation doses. Also, enzymes activities of Aspergillus terricola and
Aspergillus fumigatus were affected by gamma irradiation. α- amylase
activity decreased when gamma irradiation doses increased whereas,
cellulase and protease activities were stimulated. Gamma radiation also
affected on free amino acids contents. Gamma irradiation stimulated the
production of organic acids such as oxalic and citric acids, in the growth
medium of Aspergillus terricola and Aspergillus fumigatus as secondary
metabolites. Also, SDS-PAGE electrophoresis gave two new bands under
the influence of gamma irradiation.
