Abstract The present study aimed at producing transgenic putative lines of flax plants that may be tolerant to drought stress. The results may be summarized as follows:- 1. Biolistic transformation. Hypocotyl explants were subjected to osmotic medium for 4 hrs in the dark at 25±1°C; then ten explants were collected in the center of the plate for shooting DREB2A and GFP genes with gene gun. The bombarded hypocotyl explants were incubated at 250C for two days in the dark and the GFP expression was detected as fluorescent signals visualized using a stereomicroscope. The obtained data indicated that the mean of plate’s signal increased by the time. 2. Agrobacterium-mediated transformation Pre-cultivated hypocotyl segments were inoculated with the overnight Agrobacterium culture at different optical density (OD600 0.2, 0.4, 0.6, 0.8 and 1) for 30 minutes. Explants were co-cultivated on regeneration medium (MS medium with maltose as carbon source supplemented with 2 mg/l BA) and 100 mM acetosyringone for three days, and then transferred to selection regeneration medium supplemented with 50 mg/l hygromycin and 300 mg/l cefotax, and incubated at 25±1°C under 16 hrs photoperiod light intensity (Ca. 50 μ mol. m-2.s-1). The infected explants were subcultured every two weeks on a new selection regeneration medium. |
