Abstract

The present study aimed to isolate and characterize and comapre exosomes from she camel milk at different stages of lactation using different methods for its isolation for the first time. Milk samples were collected from adult healthy she camels at the mid and late lactation stage. Exosomes were obtained using differential ultracentrifugation and commercial exosome precipitation reagent. The morphology of isolated exosomes was examined using TEM. Exosomal marker protein (TSG101) was identified by Western blot. esRNA was extracted and major milk protein gene transcripts were detected and confirmed for its integrity and its level of expression using Real Time PCR. The concentration of phospholipid classes of exosomes (PC, PI, PS, PE) was measured using isocratic HPLC technique and compared to those obtained from cattle milk exosomes. The obtained data revealed that exosomes have been successfully isolated from she- camels milk with all characteristic of bona fade exosomes. The method of isolation had a great influence on the characteristic features of exosomes, in favor to differential ultracentrifugation in case of its availability and for limited size of samples. The stage of lactation greatly affected the expression of TSG101 protein and the concentration of phospholipid classes. Further investigations for more characterization and revealing its significance in camel milk.
