Abstract

The current study investigated whether EGCG, the major green tea polyphenol, protects normal salivary gland cells from the cytotoxic effects of 5-FU.54 adult male albino rats, weighing 150 to 200 g were selected for this study& classified into 3 groups. Group I(control group):received no treatment, Group II: the rats were administered r injection of 4mg/kg 5- FU for 5 days only, Group III: have the same protocol of group II but in combination with green tea administration from the first day till the end of the experiment at day 16th Excision of the submandibular glands was carried out and processed for histological stain with H&E &immunohistochemical stain with cleaved caspase-3(Rabbit mAb). Digital image analysis was performed for evaluation of immunohistochemical reaction. Results:I-H&E Stain Results:Group I:showed normal gland histology,Group II:showed marked atrophy of most of SA&duct system with variable degree of cytoplasmic vacuolation,Group III:showed the same histological changes of group II but with milder degree. II-Immunohistochemical results,GroupI:mild reaction was detected within the acinar cells,GroupII:showed intense immune reaction in the acinar cells&duct system,GroupIII:the acinar cells appeared with no reaction while the ductal cells revealed moderate reaction for caspase-3 at16th day.III-Statistical results:there was significant difference between all groups at the three different periods except for groupI&groupIII at the 16th day.
