Abstract Thyroid dysfunction is common endocrinal disorder, because of the thyroid gland is the master gland of metabolism and energy, any problems of the gland could affect all body systems including the oral cavity The present study was conducted to investigate the effect of thyroid dysfunction on the structure and ultrastructure of the oral mucosa of male Albino rats. Forty five adult male albino rats with 150 ±10 gr body weight were used in this investigation. The animals were divided into three groups 15 animals each. group I animals: served as controls group II animals: were used for induction of hypothyroidism using propylthiouracil which was supplied in powder form and added to the diet by through trituration in a percentage of 0.035%. group III animals: were used for the induction of hyperthyroidism using thyroid sicca which was supplied in powder form and added to the diet by through trituration in a percentage of 0.05%. The animals of the whole groups were supplied diet consisting of 50% coarse corn barley, 25% powdered sugar and 25% dry powdered milk and drinking water adlibitum, throughout the whole experimental period which lasted for three months. Summary and conclusion 260 At the end of the experiment the animals were sacrificed by cervical dislocation, their tongues, and samples from the gingiva (from the region of the first molar) were taken out and were prepared to be examined through: - Microscopic and submicroscopic examination to detect any structural and ultrastructural changes in the surface epithelium and lamina propria of the lingual and gingival mucosae. - Scanning electron microscope of tongue to elucidate the surface ultrastructural changes in the tongue papillae - Histochemical demonstration of acid mucopolysaccharids in the mucosae under investigation. - The universal dakocytomation labeled streptavidin – biotin 2 system horseradish peroxidase enzyme with PCNA mouse monoclonal antibody for localization of PCNA in the aforementioned structures. PCNA labeling index was counted and treated statistically. - The ultravesion mouse tissue detection system using antimouse monoclonal antibody for demonstration of E-cadherin to detect any possible changes Thyroid glands were also taken out, fixed, processed, sectioned and stained with hemotoxylin and eosin for histological evaluation |
