Abstract

The present study was conducted to evaluate the toxopathological effects of AFB1 on pregnant female rats and their offspring. Twenty eight female Wistar rats obtained from the Laboratory of Animal Resource, Cairo, with an average age 2.5-3 monthswere used in the present study.They were divided into four equal groups,group 1& 2 were kept as control groups, where group 2 was administered the vehicle (corn oil). Group 3 &group4 were administered AFB1 at dose of 1mg/kg. b. wt. from 6-15 days of gestation period, Group 3 was sacrificed at 20th day of gestation period, and group 4 left to normally parturate and sacrificed three weeks post-partum.
Group 3 and group 4 were investigated for clinical signs, histopathological, teratogenic and genotoxic effects of AFB1 in both dams and feti.In addition observation of the feti which were aborted and those which was born alive.
The obtained results of dams were significant decrease in the body weight, pathological investigations revealed gross enlargement in the size of liver, congestion and other pale yellow discoloration areas. mild congestion, red hepatization and firmness of the lung,histopathologically the liver show individual cell necrosis, disorganized hepatic cord pattern and the central vein is dilated and congested,kidney revealed interstitial hemorrhages, cast in their lumens, degeneration and necrosis of tubular epithelial cells,lung revealed lymphocytic infiltrates in the interstitial tissue and the spleen revealed lymphoid depletion in the white pulp and hemosidrosis in the red pulp.
Chromosomal analysis revealedboth structural and numerical chromosomal aberrations.Structural chromosomal aberrations recorded were chromatid breaks, chromatid gaps, Centromeric attenuation, end to end association, fragment, ring chromosomes, dicentric chromosomes, chromosomal fusion, centric fusion, stickness, deletions and fragments.
Studies on feti of group 3 revealed significant decreased mean fetal weights and fetal crown to rump lengths, wrinkling of skin, adactyliaand exophthalmia. Histologically dissociation of the hepatic cords, central vein and hepatic sinusoids are congested and engorged with RBCs, Kidney revealed degenerative changes in the renal tubules, degeneration and atrophy of the glomeruli, thymus revealed alterations characterized by lymphoid depletion,skeletal anomalieswereobserved and consisted of incomplete ossification ofskull bones and failure of ossification of small bones.
Group four revealed storm of abortion of some treated rats and the feti which was born alive were weak and show stunted growth with lower growth rate and was dead after 2 weeks after parturition
