Abstract

The present study aims to assess the anticonvulsant potential of HUCB-derived mesenchymal stem cells in PTZ induced kindling in rats and to compare their effects with the anticonvulsant drug gabapentin.
In the present study 30 male albino Wister rats were used and were divided into the following five groups:
Group (1): Normal control rats.
Group (2): Saline control group; received 0.5 ml saline by I.P. injection, parallel to PTZ injection.
Group (3): Disease control group; subjected to PTZ (30 mg/kg I.P.) three times a week, for a total of 13 injections and left without treatment.
Group (4): (Gabapentin treated group); kindled rats that received I.P. gabapentin 60 minutes before each PTZ injection at the dose of 200 mg/kg.
Group (5): (Mesenchymal stem cell treated group); kindled rats that received I.V. HUCB-derived mesenchymal cells at a dose of 1x 106 cells/rat in the tail vein beginning after 22 days of PTZ injection (after the 10th injection) as a single dose.
After each PTZ injection rats were observed for 30 minutes to record modified Racine score of epilepsy then rotarode test and open field tests were performed to assess motor coordination. Finally after the 13th injection all rats were sacrificed and their brains were obtained for the following;
1- Tests for biochemical markers of oxidative stress
2- Immunohistochemical tests for anti GABA A antibodies
3- Immunohistochemical tests for S100 marker
The results obtained could be summarized as follows:
1- Administration of PTZ caused an increase in modified Racine score, a decrease in the latency to fall time from the rotarode apparatus, a decrease in the number of squares crossed and activity factor in open field test, increase in MDA level and a decrease in GPX, SOD and catalase activities.
2- Administration of Gabapentin induced a decrease in modified Racine score, increase in the latency to fall time in rotarode apparatus, increase in the number of squares crossed and activity factor in open field test, a decrease in MDA level, no improvement in GPx activity and an increase in SOD and catalase activities.
3- Administration of mesenchymal stem cells resulted in a decrease in modified Racine score, increase in the latency to fall time on rotarode apparatus, increase in the number of squares crossed and activity factor in open field test, decrease in MDA level and increase in GPX, SOD and catalase activities.
4- Immunohistochemical results suggest that mesenchymal stem cells may act through increasing the level of the inhibitory amino acid GABA.
So it is concluded that:
Mesenchymal stem cells had a therapeutic role in chronic epilepsy may be through increasing the level of inhibitory amino acid GABA
