Abstract

Milk is a complex biological fluid that contains a wide variety of constituents and possesses unique physical and chemical characteristics. In addition of being nutritional medium,it also presents a favorable physical environment for the multiplication of micro organisms. Milk may serve as a vehicle for great varieties of food borne pathogens, especially staphylococcus aureus as one of the most important pathogen in milk.Staphylococcus aureus is ubiquitous in nature and its major habitats include the skin and mucous membrane of the nose .Also, lactating animals and human handlers are main sources for this bacterium and frequently implicated in the transmission of this pathogen.Although staphylococcus are not sufficient cause to incriminate a specific food Poisoning as they are not all enterotoxigenic , Yet ,different types of enterotoxins (SEs)are produced by some S. aureus strains . Once formed, SEs are extremely difficult to eliminate from foods as they are resistant to heat, freezing and irradiation .Also, SEs may survive commercial pasteurization processes .Moreover, they are highly stable and resist most proteolytic enzyme.Most staphylococcus isolates produce toxins are responsible for staphylococcal food poisoning outbreaks;accordingly, the presence of S.aureus in food is considered a potential health risk.Therefore, this study was designed to throw spot lights upon S.aureus isolated from raw (buffalo) milk, studingit’scultural, biochemical and In-vitro anti-microbial sensitivity for it with special reference to some virulence genes.So the present study was performed on a total of 100 milk sample from small scale producer s shop and market at El-Menofyia Governorate.The incidence of staphylococci in the examined samples is (80%).Incidence of coagulase positive staphylococci is 42 represent (53%) while incidence of coagulase negative is 38 represents (47%).Incidence of staph aureus is 31 represent 73.8% .The results of anti-biotic sensitivity tests for S. aureus revealed that Gentamycin, Trimethoprim / sulphamethazole , Ampicillin, Cephradine, were the most proper antibiotics with the highest in vitro efficiency against isolated S. aureus but they were resistant to Flumox, Rifamycin, Amoxicillin and Clindamycin.PCR results for S.aureus showed that spa virulence gene was detected in 1 studied strain (16.6%) out of 6 isolates , while see virulance gene was detected in 2 studied strain (33.3%) out of 6 isolates.
