Abstract

A total number of 56 male Albino rats (divided into 7 groups each consisted of 3 control and five treated were used for studying the sequential cardiotoxic effect of DOX at different post treatment periods using intraperitoneal cumulative dose which is similar to that used in human (10 mg/kg body weight). A single 2 mg/kg body weight of DOX is cardiotoxic in rats. It produced acute toxic myocarditis characterized by degenerative and necrotic changes in myocytes, mitochondrial damage and vacuolation, vascular changes and occasionally inflammatory cells infiltration. These changes increase in severity with subsequent treatment with the same dose till reaching to a cumulative dose of 6 mg/kg body weight during a period of 15 days. There after a cumulative dose of 8 mg/kg body weight with a withdrawal period of 15 days or a cumulative dose of 10 mg/kg body weight (therapeutic cumulative dose in man) with withdrawal period of two or three months produced predominant changes of chronicity such as disorganization and atrophy of myocytes, condensation and atrophy of mitochondria and fibrosis with occasional focal necrosis indicating incomplete elimination of DOX and/or its metabolites.
