Abstract This study was conducted to examine the association between HPV and EBV with oral squamous cell carcinoma and ameloblastoma assessed by detection of their DNA in these tissues and to correlate the presence of the viral DNA and aggressiveness of these tumors. Thirty formalin fixedand paraffin embedded blocks was used in the study.Fifteen of them were previously diagnosed as oral squamous cell carcinoma and fifteen of them were previously diagnosed as Ameloblastoma. Using hematoxylin and eosin stain, reevaluation of these cases was carried out to confirm the diagnosis and establish the histopathologic grading. According to the histopathological criteria, squamous cell carcinoma cases were categorized as 5 well differentiated,7 moderately differentiated and 3 poorly differentiated squamous cell carcinoma. While Ameloblastoma cases were categorized as 11 benign cases and 4 malignant cases. DNA was extracted from formalin fixed, paraffin-embedded tissues using The DNeasy Blood &Tissue Kit (QIAGEN),The Light-Cycler instrument (Roche –Germany) permits the simultaneous amplification and quantification of the PCR-product at each cycle. And we used the SYBR Green system for the detection of EBV and HPV. |
