Abstract

Diabetes causes microvascular and macrovascular complications. Cardiovascular complications of diabetes are the primary cause of morbidity and mortality among patients with diabetes and accounts for more than 80% of deaths. This work was conducted to investigate effect of MR blockade on the diabetes-associated cardiovascular complication and whether the combined therapy of spironolactone with enalapril can provide an additional beneficial effect compared to each treatment alone. In addition, we attempted to elucidate the underlying mechanism(s) involved in this setting. The protective effects of either of SPIRO and ENAL were reassembly mediated by the decline in heart weight index, cardiac isoenzymes, MDA level, NOX2 mRNA expression, iNOS protein expression, TGF-β protein expression and MR mRNA expression. Moreover, both drugs could reverse the decline of antioxidant enzyme activities and eNOS protein expression. MR blockade may provide an additional treatment option for the management of cardiovascular complication in patients with diabetes.
