Abstract This study aimed to : Osteoarthritis is highly prevalent chronic disease affecting synovial joints leading to irreversible damage to all joint structures. OA develops due to multifactorial disorders and divided into primary and secondary OA. The current options for the treatment of OA mainly help to reduce the symptoms of pain and inflammation with neither effect on modification nor slowing progression of joint structural changes. Many experimental trials and preclinical evaluation are carried out to find a promising OA disease modifying drugs, among these drugs are statins. Aim : This study was conducted to evaluate and compare possible prophylactic and therapeutic effects of oral, interarticular or combined oral and interarticular atorvastatin administration in experimental model of osteoarthritis as a potential OA disease modifying drug. Setting : Medical experimental research center, Faculty of Medicine, Mansoura University. Results : Atorvastatin was found to have beneficial effects in OA demonstrated by decrease inflammatory cytokines (IL-1, TNF), proteolytic enzyme (MMP-13), oxidative stress NO and MDA, increase in GSH and improvement in histological examination in all treated groups and the improvement was more evident in combined groups either prophylactic or therapeutic and the beneficial effects of atorvastatin were statistically more significant in prophylactic group as compared to corresponding therapeutic group. Conclusion : Atorvastatin was found to have beneficial effects Therefore, it is advised to administrate atorvastatin in OA patient and the earlier the atorvastatin treatment the better the improvement and outcome as demonstrated in this experimental study. |
