Abstract

Acrylamide is known to cause cancer in experimental animals. Commercially, it is used in the manufacture of some plastics as well as various other materials.In search for new substances, which might act as anticancer agents, this study was conducted to investigate the effect of treatment with Punica granatum, Psidium guajava extracts as well as vitamin E on physiological, biochemical and histopathological changes in rats treated with acrylamide.
For this purpose, thirty-six male Sprague-Dawley rats of same age and body weight were selected and divided randomly into six equal groups: control (CG), Acrylamide (AA), Pomegranates peel extract + Acrylamide (PPE+AA), Guava leaves extract +Acrylamide (GLE+AA), mixture of pomegranates peel and guave leaves extracts +Acrylamide (PPE& GLE +AA) and Vitamin E +Acrylamide (Vit.E+AA). AA, PPE, GLE and Vit. E were given by gavage at doses 50, 200, 200, 100 mg / kg body weight respectively. The blood and tissue samples were taken from all rats on the 35th day to be analyzed for the determination of physiological, biochemical and histopathological changes.
Regarding physiological parameters, the results showed that that the daily body weight gain of rats in groups AA, PPE+AA and PPE&GLE+AA were significantly decreased when compared with the normal control group (CG). On the other hand, rats administrated with Vit.E showed no significant increase in the daily body weight gain (3.4 g) as compared with rate administrated only with acrylamide (3.83 g) concerning of the daily intake
Some biochemical changes were observed in blood serum of rats treated with acrylamide, Pomegranates peel extract, guave leaves extract and vit. E.Assay for serum AST and ALT activities, showed that the levels of AST and ALT were significantly higher (~2.5 and 1.5 respectively) in AA treated group than the control rats. AST and ALT cytosolic enzymes of hepatocyte and as an increase of their activities reflect on increase in plasmic membrane permeability, which in turn is associated with cell death. By applying treatments PPE, GLE, PPE + GLE and VitE the AST activity under respectively studying showed values of 24.39, 24.26, 24.82 and 23.17 (IU/ L). The
respective values of ALT are 23.72, 23.03, 23.53 and 22.48 (IU/ L). It is quiet that the treatment with PP or GL or mixture of their extracts as well as with vitamin E have a remarkable protein of serum aspartate amino transferase (AST) and alanin amino transferase (ALT) levels toward acylamide induced hepatoxicity.
