Abstract

Our study evaluate the effect of Flavocoxid and Allicin on atherosclerosis induced in rabbit by high cholesterol diet. The following inflammatory and oxidative stress markers are measured: Triglyceride, total cholesterol, low density lipoprot Lipid peroxidation lipid (MDA), reduced glutathione and level of superoxide dismutase C- reactive protein Aortic rings were isolated to evaluate endothelium-dependent vasorelaxation. Aortic expression of level of TNFα and NF-κB was determined immunohistochemically. The Study revealed that both allicin and flavocoxid significantly improved vascular reactivity, and increased serum HDL level. However they did not significantly change serum TC, TGs and LDL-C levels. Both flavocoxid and allicin significantly increased GSH and SOD, decreased MDA and decreased aortic TNFα and NFκB expression. The present study was also designed to evaluate whether allicin and flavocoxid could have anti-inflammatory and/or antitumor activity in vitro and invivo against Ehrlich ascites carcinoma cells (EAC). The study showed that: 1.Flavocoxid (but not allicin) has the ability to enhance percent survival in mice compared to cisplatin treated group 2.Both allicin and flavocoxid have ability to reduce tumor size in mice compared to untreated group showing a similar pattern as cisplatin-treated group.3. Allicin was superior to both flavocoxid and cisplatin in terms of reducing inflammation and necrosis scores 4. Both allicin and flavocoxid had the ability to reduce TNF-α and NFκβ expression when compared to EAC group.
