Abstract

The present work was planned to study the deleterious effect of prenatal administration of a benzodiazepine derivative, flunitrazepam (Rohypnol), on the pregnant albino rats and the reflection of any impairments on their foetuses. Flunitrazepam was administered orally with the therapeutic dose (0.18 mg/kg b.wt.) and double of the therapeutic dose (0.36 mg/kg b.wt.) from

day of gestation (period of organogenesis).
On the 20th day of gestation rohypnol treated mothers have revealed decrease in their body weight and lethargy in their movements. In addition increased percentage of abortion and decrease in maternal uterine weights were observed.
The effect of rohypnol on the foetuses were characterized by lethargy, growth retardation, high percentage of resorption, haematoma, paralysis in both limbs, talipomanus and hind limb loss. Incomplete cleft palate with arched palate were also seen in foetuses of high dose treated group (0.36 mg), while arched palate only was found in those of low treated group (0.18 mg).
from histological points, the liver, kidney, lung and brain of the pregnant rats and their foetuses showed many histological changes. These changes were dose-dependent.
