Abstract

Drug induced gingival overgrowth (DIGO) is known as an adverse effect with three types of: PNT, anti-epileptic drug; cyclosporine A (CSA), an immunosuppressive drug; and calcium blocker. Aim of the work : This study was carried out to identify and outline the possible biological roles of KGF and CTGF in PNT and CsA induced gingival hyperplasia. This study was carried out on sixty adult male albino rats were divided into 3 groups : group I : (control group). The rats of this group will receive no treatment. group II : The rats of this group will be administrated CsA in mineral oil via gastric feeding daily for 7 weeks. group III : The rats of this group will be administrated PNT after trituration with tween 80 solutions they received PNT via subcutaneous Twice daily for 6 weeks. At the end of the experiment, all the rats will be sacrificed and their gingivae will be excised .Routine tissue processing will be carried out for staining with: - Hematoxylin and eosin stain. - Immunohistochemical staining for KGF and CTGF will performed according to the manufacture’s instruction. The Immunohistochemical staining will be assessed and scored. The results of this study will be analyzed statistically to detect the possible statistical significant differences and correlation between the different variables. Results: 1. Hematoxylin and Eosin. a- Degree of gingival overgrowth. Control group exhibited score zero GO while group II yielded score 2 and score 3with blunt and bulbous gingival crest mean while rats of group III revealed less pronounced gingival hyperplasia and gingival crest was knife edge. b- Histological findings. group I revealed the feature and a of architecture of rats gingiva while groups II and III showed GO with variable extents. 2-Immunohistochemical findings KGF. group II had the highest mean value while control group showed the lowest one statistical, these was an overall significant difference between the studied groups. As well as between each two groups. CTGF group II had the highest mean value while control group revealed the lowest one. Statistical analysis revealed an overall significant difference between the studied groups and between each two groups. Regarding correlation between KGF and CTGF for all groups, Pearson correlation test yielded a non-significant association between the two antibodies in all groups.
