Abstract The present work was carried out for genetic evaluation of 23 somaclones and gametoclones derived from in vitro tissue culture of 4 commercial Egyptian bread wheat (Triticum aestivum L.) cultivars. All 27 genotypes were subjected to agronomical, cytological, tissue culture and DNA molecular marker studies. Original parental wheat cultivars were used as control for their derived clones. As general remarks, this study revealed that the application of plant tissue culture techniques may be able to produce useful genetic variation in wheat and this variation lead to superior genotypes than their parental cultivars in high yield. Those genotypes are promising for increasing the production of wheat in Egypt. Measuring the cytological stability of gametoclones, somaclones and comparing with their parental cultivar from time to time is very important in wheat program. Moreover, the present study detected many DNA bands that can be used for distinguish between the different genotypes, which will help to speed up the breeding programs. |
