Abstract Development and Application of Non-Parametric and Robust Linear Regression Methods for the Analysis of Some Nutraceuticals and Dietary Supplements The thesis comprises three main parts: Part I This part is mainly concerned with the introduction of the novel term ―Nutraceuticals‖; its definition, history of nomenclature and the use of food as medicine. It gives a detailed classification of the broad title ―Nutraceuticals‖ into subtitles of Dietary Supplements, Functional Food, Medical Food, and Farmaceuticals. A brief of market and demand as well as regulations of nutraceutical products is also included. The part then specifies the studied nutraceuticals throughout the thesis, their chemical names, structures, physical and pharmacological properties. It also involves literature reviews for the reported classical and chemometric assays of the selected drugs in pharmaceutical dosage forms as well as in biological fluids. Part II Single Component Analysis This part is divided into two chapters: Chapter I deals with two validated spectrophotometric and spectrofluorimetric methods for the determination of Alpha Lipoic Acid (LA) in bulk drug and capsules using merbromin reagent. This chapters starts with an introduction for the types of fluorescence quenching; Static quenching and dynamic quenching. It points out the sequential equations relating the fluorescence quenching linearily versus the corresponding drug concentration. A typical plot of Stern-Volmer equation is represented. A literature review of the use of merbromin reagent and its related compounds in spectrometric determination of pharmaceutical compounds is included. This extends to the spectrophotometric determination of drugs through complexation with merbromin as well as the use of fluorescence quenching of Merbromin and related compounds in drug analysis. Two simple and sensitive analytical methods are then described in detail for the determination of Alpha Lipoic Acid (LA) in bulk drug and its pharmaceutical dosage form. The first method (I) is spectrophotometric determination of the chromogen formed between LA and Merbromin (MER1) in an aqueous buffered medium. This may be attributed to the coordination of sulphur center with MER1 (merbromin of 40 mg%, prepared for colorimetric method). A red coloured complex, that exhibits an absorption maximum at 548 nm, is produced. The different experimental parameters affecting the colour development and its stability were studied and optimized. They have been optimized to achieve high sensitivity, low blank readings and high stability of the formed complex. |
