Abstract

The present experiments were carried out at the Microbial Biotechnology Department and other labs in Genetic Engineering and Biotechnology Research Institute, Sadat City University, Sadat city, Egypt. The main objective of the present investigation was the production purification and characterization of microbial cellulose hydrolyzing enzyme. This study was carried out as following:- 1) Isolation of alkaliphilic bacteria that hydrolyzed the cellulose based polysaccharide. 2) Purification and characterization of bacterial isolates. 3) Purification and characterization of alkaliphilic cellulose hydrolyzing enzyme. The first experiment was the screening for cellulosic activity of bacteria isolated from soils at ―Menia El-Qamh, Sharkiya government and Sadat City, Menofiya government, Egypt ”. The result was 24 positive isolates recognized that hydrolysis of CMC and three isolates marked MK7, MK16 and MK28 were found to be the most potent isolates. The second experiment was the cloning, subcloning and sequencing of cellulase encoding gene. The third experiment was the production and purification of cellulose hydrolyzing enzyme using affinity chromatography.
