Abstract The objective of this thesis to prepare some topical formulations containing Ketorolac Tromethamine (KT) niosomes to overcome problems associated with KT as short half-life and gastric irritation. Thus the work in this thesis was divided into five chapters: Chapter I: Preparation and characterization of KT loaded niosomes: Preparation of different formulae of KT niosomes using ether injection method. A Box-Behnken design was selected for formulating ketorolac tromethamine niosomes. Chapter II: In-vitro release and kinetic studies of KT loaded niosomes Determination of the in-vitro release of the drug from the prepared niosomes. The kinetic parameters for the in-vitro release data were determined. Chapter III: Factorial characterization of KT loaded niosomes: Correlation between the independent variables (X1, X2 and X3) and the dependent variables (Y1, Y2 and Y3). characterization of ketorolac tromethamine niosomes using response surface methodology. Chapter IV: Preparation and evaluation of KT loaded niosomes from some topical dosage forms: Preparation of KT niosomes in different topical formulations. The in-vitro release studies, kinetic order determination and the viscosity of the prepared topical formulations were determined. Chapter V: Clinical evaluation of the anti-inflammatory activity of topical formulation containing KT loaded niosomes. Evaluation of the anti-inflammatory activity of KT loaded niosomes from the selected topical formulations using carrageenan induced rat hind paw edema method. |
