Background:
Several candidate genes have been identified they all encode cytokines involved in the regulation of IgE synthesis.
Aims:
The present study aimed at studying variant allelic changes in IL4 receptor - chain gene on chromosome 16p12 and their relation to bronchial asthma.
Methods:
This study comprised 25 children with atopic asthma, 15 males and
10 females. Their ages ranged from 4 to 13 years. 10 normal healthy non‐atopic age matched volunteers were chosen as controls. They included 6 males and 4 females, their ages ranged from 8 to 12 years. All of them had no past or present history of any other chest or systemic disease. They had neither systemic nor topical corticosteroid therapy at least two weeks before the study and no systemic antihistamines at least seven days before the study Both patients and control groups were subjected to the following:
DNA extraction: Peripheral blood leukocytes were collected on 0.5 M EDTA as anticoagulant. DNA
extraction was done using alkaline lysis based technique .
Polymerase Chain Reaction (PCR) Primers were produced as dried oligonucleotides, Taq DNA polymerase enzyme and Nucleotides 10 mm Blood samples prepared from whole blood from studied groups.
Results:
18 homozygotes for ile50/ile50 allele and 5 controls.
3 heterozygotes for ile50/val50 and 2 controls.
4 homozygotes for val50/val50 and 3 controls.
Conclusion:
Polymorphism and variation of ile50 (isoleucine) 50 /val50
