Abstract This study was conducted to use the tissue culture techniques in order to propagate goldenrod (Solidago canadensis) through estimating the most convenient concentrations of applied plant growth regulators to achieve the best micropropagation, protocol. Shoot tips and nodal segments were used as explants. This work was conducted at the plant tissue culture laboratory of the Floriculture and Landscape Gardening Department, Faculty of Agriculture, Alexandria University, during both seasons of 2014 and 2015. The present study included three major experiments; the first experiment was to find the best way for surface sterilization of goldenrod explants (shoot tips and nodal segments). Mercuric chloride (HgCl2) was used at a concentration of 1 g/l for different durations (4, 6 and 10 min.) in a combination with ethyl alcohol 70% for different durations (6, 30 and 60 sec.). |
