Abstract

The present study was conducted to evaluate the potential hepatotoxic effect of 5- HMF in mice through measuring the liver function biomarkers, hepatic antioxidant assay, genotoxic biomarkers (8-hydroxy-2-deoxyguanosine and DNA fragmentation), in addition to histopathological investigation of liver tissue and the immunoreactivity of the apoptotic regulating protein BCL-2.
The present study also aimed at assessing the ameliorative effect of EVOO against HMF induced hepatotoxicity.
For this purpose, 80 mice were assigned into four equal groups each containing 20 mice. group one kept as control with intraperitoneal injection by 0.5 ml/kg b.wt of 0.9 NaCl three times weekly for 4 weeks, group two was given 5-HMF (250 mg/kg in 0.9 % NaCl) by intrapertoneal injection 3 times weekly for 4 weeks. group three (5-HMF exactly as previous + Olive oil with dose of 300 ul/kg orally daily for 4 weeks and group 4 received Olive oil exactly as group 3.
At the end of the examination the mice were relinquished, blood was collected without anticoagulant then centrifuged at 3000 rpm for 10 minutes for division of serum then put away at - 20°C for chemical investigation. Liver was separated into three Parts. The first part stored at (-20°C) for determination of antioxidant indices. The second part was preserved at (- 80 oC) for comet assay. The third part was fixed in 10% neutral buffered formalin for histopathological and immunohistochemical examination.
