Abstract

Coffee is one of the most popular and widely consumed beverages in the world. Coffee consumption have beneficial effects on human health because coffee contains several hundred chemical compounds including flavonoids, phenolic, caffeine and chlorogenic acid so the coffee used for traditional remedy, and as traditional herbal medicine, especially for obesity. Different kinds of coffee (dark, light and green) were analyzed for it is chemical constituent, antioxidant activity, total phenolic and total flavonoids then added in different levels (2-4 ml) to forty albino obese rats drinking to improve their lipid profile compared to negative control group (5 rats). All rats were fed on basal diet for one week for adaptation, then the rats were divided into two main group: The first main group (n = 6): weighting 120±5 g rats, were fed on the basal diet as negative control. The second main group (n = 42): weighing 220±20 g (obese rats). The obese rats were divided randomly into seven subgroups (6 rats each) according to the following Subgroup (1) Rats were fed on the basal diet as a positive control. Subgroup (2): Rats were fed on the basal diet and were given oral injection 2 ml of dark coffee. Subgroup (3): Rats were fed on the basal diet and were given oral injection 4 ml of dark coffee. Subgroup (4): Rats were fed on the basal diet and were given oral injection 2 ml of green coffee. Subgroup (5): Rats were fed on the basal diet and were given oral injection 4 ml of green coffee. Subgroup (6): Rats were fed on the basal diet and were given oral injection 2 ml of light coffee. Subgroup (7): Rats were fed on the basal diet and were given oral injection 4 ml of light coffee. At the end of the experiment (28 days) rats sacrificed and internal weight (liver, kidney, spleen, heart and lunges) were recorded. Rats were weighted at the beginning and the end of the experiment to determined body weight gain (%). Blood samples were collected to determine lipid profile, liver function, kidney function and antioxidant activity in blood serum. The obtained results can be summarized as follows: 1- The different kinds of coffee (green, dark and light) contained .57% flavonoids, 12.8% phenolic, 5.93% caffeine and 92% antioxidant activity. 2- interestingly, dark coffee had the highest caffeine content (7.02%). 3- Increased blood fat and cholesterol levels and weights as a result of obesity was corrected these percentages when giving the rats different kinds of coffee. 4- There were improved in total cholesterol and triglycerides in obese rats which administrated with dark, light and green coffee . 5- group which administrated with 4 ml of dark coffee was more effective in decrease TL , LDL and VLDL than rats which administrated with 2 ml of dark coffee. 6- Administrated rats with 4 ml of dark coffee was more effective in reducing AST, ALT, ALP and uric acid than all groups. 7- Obese rats which administrated with 2 and 4 ml of dark coffee increased GTF and catalase by 1.43%, 7%, 12.15% , 43.76% while decrease MDA by 25.86% and 21.55% respectivelly 8- There were no significant differences in all organ weights between rats which administrated with 2 and 4 ml of dark coffee. 9- Administrated rats with 2 and 4 ml of light coffee decrease TL, TC, TG, LDL and VLDL by 7.14, 33.8, 12.5, 25.9, 15.3% and 5.2, 22.1, 8.3, 28.5 and 10.4% resepectivelly. 10- Administrated rats with 2 and 4 ml of light coffee increase GTF and catalase by 32.4%, 24.6% 35.9% and 38.9% respectively 11- group which taken 2 and 4 ml of light coffee had the same effect on liver and kidney functions 12- Administrated rats with 4 ml of light coffee was more effective than administrated with 2 ml of light coffee in reducing final weight .13- There were no significant between rats that administrated with 2 and 4 ml of light coffee in liver, spleen, heart and lunge weights. When administrated rats with 4 ml of light coffee was more effective in reducing kidney weights than other obese rats. 14- Rats which administrated with 4 ml of green coffee had lower in TC and LDL than rats administrated with 2 ml of green coffee. 15- Administrated rats with 2 and 4 ml of green coffee increase GTF and catalase by 20 , 34.6% and 25.4 and 32.7% respectively. While MDA had opposite trend. 16- The highest reduction in ALP, urea and creatinin was observed in rats administrated with 4 ml of green coffee than all obese groups. 17- Administrated rats with 4 ml of green coffee was more effective than administrated rats with 2 ml of green coffee in reducing final weight and body weight gain. 18- Rats which administrated with 4 ml of green coffee was more effective than rats administrated with 2 ml of green coffee in reducing liver, spleen, heart, kidney and lunges weights.
