Abstract

Periodontitis is bacterially mediated, chronic inflammatory disease of the tooth-supporting tissues. Destruction of extracellular matrix by MMPs is a main event in periodontitis. Inflammatory responses were shown to be regulated by several factors including epigenetic factors. So, this study was to investigate if T. denticola a periodontal pathogen could regulate MMP-2 expression and activation in PDL cells via epigenetic modifications. PDL cells were challenged with T. denticola for 2 hours then incubated for specific periods. MMP-2 activity and fibronectin fragmentation were then investigated. Transcription of “MMP-2, MT 1-MMP, and TIMP-2” were also investigated by PCR. PDL cells challenged with T. denticola showed higher expression and activation of MMP-2, MT 1-MMP, and TIMP-2 and fragmentation than control cells at different time points. T. denticola modified the PDL cell behavior into an abnormal destructive manner that, to some extent, similar to the chronic nature of periodontal diseases. The change of the transcriptional levels suggests that T. denticola caused epigenetic modifications in these genes.
