Abstract

In vitro isolation and characterization of mesenchymal stem cells derived from ovarian surface epithelium (OSE) cells. In vivo to test the hypothesis that the treatment with obtained mesenchymal stem cells of OSE origin could improve premature ovarian failure (POF) induced in mice by chemotherapy intraperitoneal injection of cyclophosphamide.This study was conducted on Seventy (70)female C57BL/6 mice aged (6) weeks and weighing about 20-23 g (obtained from animal facility of the Experimental Medical Research Center (EMRC), Faculty of Medicine, Mansoura University, Egypt). To prepare recipients, wild-type female was received 120 mg/kg cyclophosphamide which induced damage to the primordial follicles cohort in the ovary. The ovaries were obtained under pentobarbital anesthesia and examined at 48h, 72h, 96h, 2weeks and 4weeks to evaluate the chronic phase of ovarian damage and verify for the self-healing process. Mice were divided into 4 groups. group (I) (negative control each contain 25 mice). group (II) (POF cyclophosphamide induction each contain 25 mice). group (III) (cyclophosphamide treated with DMEM culture media alone each contain 10 mice). group (IV) (cyclophosphamide with stem cells treated each contain 10 mice). Five mice were sacrificed at different intervals in group I & II after 48h, 72h, 96h, 2weeks and 4 weeks for obtaining ovarian tissue and blood samples.For group III & IVFive mice were sacrificed after 2weeks and 4 weeks for obtaining ovarian tissue and blood samples. Clotted blood samples were collected for biochemical investigation.OSE stem cells after isolation from the surface epithelium obtained was characteristics by their adherence to plastic consisted of heterogeneous cell population.The results of the flow cytometry analysis for mice ovarian surface epithelium (OSE) mesenchymal stem cells showed that there was positive expression for surface antigen CD14, CD29, CD44 and Oct4 (passage 3) . and negative expression of surface antigen CD45 and CD34. There was highly significant decrease in serum estradiol (pg/ml) of cyclophosphamide vehicle-treated group (POF) at 24h, 72h, 96h, 2 weeks and 4 weeks, in addition, there was insignificant difference in estradiol of (POF) in the stem cells-treated group after 2 weeks and 4 weeks compared with control group. There was highly significant increase in FSH (mIU/ml) of (POF) mice after 24h, 72h, 96h, 2 weeks and 4 weeks in cyclophosphamide in the vehicle-treated group ,moreover,there was insignificant difference in FSH of (POF) mice in the stem cells-treated group after 2 weeks and 4 weeks compared to that of control group. There was highly significant decrease in serum progesterone (ng/ml) in the of (POF) mice after 24h, 72h, 96h, 2 weeks and 4 weeks in cyclophosphamide in the vehicle-treated group , moreover, there was insignificant difference in the in the progesterone of (POF) mice in the stem cells-treated group after 2 weeks and 4 weeks compared with control group.The data obtained from histopathological examinationshowed that all experimental studied groups have primordial, primary, secondary and atretic , but there is a significant change in the degree of each primordial, primary, secondary and atretic, in the statistical results between groups was observ Mesenchymal stem cells of OSE origin could improve premature ovarian failure (POF) induced in mice by chemotherapy intraperitoneal injection of cyclophosphamide.
